Medium optimization can enhance microbial growth and increase product yield.Optimization experiments often begin with classical One-Factor-at-a-Time studies or with statistical screening designs such as Plackett&#8211;Burman.The one-factor-at-a-time method adjusts one variable, such as pH, while keeping the others constant.But multiple biological variables often interact, so changing pH, for example, can change how nutrient levels affect growth. As the number of variables increases, this method becomes inefficient.The Plackett-Burman design can simultaneously screen and identify the most influential variables.Each variable is tested at high and low levels, such as pH 9 and 6 or carbon concentrations of 10 and 5 grams per liter.Each run uses a unique combination of factor levels, helping to understand which factors most affect the result. For example, higher pH or an increased carbon source may enhance protein yield.In a standard Plackett&#8211;Burman design, N runs can screen up to N&#8722;1 variables, with N typically chosen as a multiple of four&#8212;for example, 8 runs can screen up to 7 variables.

This method employs a matrix format, where each variable is tested at two levels&#8212;typically high (+) and low (&#8722;). Each experimental run features a unique combination of high and low levels across variables, enabling the estimation of the effects of each factor on the response.The number of runs in a Plackett&#8211;Burman design is typically a multiple of four, and the design can accommodate up to N&#8722;1 variables in N runs. This screening approach helps focus experimental resources on the most influential variables, making it well-suited for preliminary optimization. Microbial growth media can be improved by testing which ingredients most affect growth and product yield. Statistical experimental design gives a structured way to do this. It also makes the process more reproducible and efficient.

The One-Factor-at-a-Time, or OFAT, method changes one variable while all others stay the same. This approach is simple, but it cannot show interactions between variables. When two factors work together, OFAT may miss that effect and lead to a less effective result.

The Plackett-Burman design, or PBD, screens many variables at once. It uses a matrix where each factor is tested at two levels, usually high (+) and low (−). Each run combines these levels in a different pattern, which helps estimate the effect of each factor on the response.

PBD is useful when many variables need to be checked with few experiments. The number of runs is usually a multiple of four, and an N-run design can test up to N−1 variables. It is orthogonal for main effects, so each main effect can be estimated independently of the others. However, the results may still include factor-factor interaction effects, so follow-up tests are often needed. For example, 8 runs can screen up to 7 variables, which makes PBD a strong choice for preliminary screening rather than detailed optimization.

After the most important factors are found, the Taguchi method or Response Surface Methodology, or RSM, can be used to refine their levels. The Taguchi method uses orthogonal arrays and signal-to-noise ratio analysis to reduce variability and improve quality. RSM uses statistical models, often second-order polynomials such as Central Composite or Box-Behnken designs, to study interactions and curvature in the response. It can also generate a predictive response surface that helps visualize the best conditions.

Together, these methods support a data-driven approach to media development. They help match microbial performance with industrial production goals.
