Q1: How do you measure blood pressure using a sphygmomanometer?
Place the sphygmomanometer cuff around the subject's arm and insert stethoscope earpieces into your ears. Position the diaphragm under the cuff on the inside of the elbow. Pump the cuff to 200 mm of mercury, then slowly release air by turning the metal knob. The first Korotkoff sound heard indicates systolic pressure, while the last sound signifies diastolic pressure.
Q2: Why does body position affect systolic blood pressure measurements?
The experimental hypothesis predicts that systolic blood pressure will be higher when sitting compared to lying down because more pressure is required for blood to be moved around the body against gravity. This positional difference reflects how the circulatory system adjusts to maintain adequate blood flow in different body orientations.
Q3: What is the operculum and how does it relate to fish respiration?
The operculum is a bony flap covering the gills, located behind and below the fish's eye. When the fish respires, the operculum moves in and out, and each movement counts as one gill beat. Counting gill beats provides a direct measurement of the fish's respiration rate during the experiment.
Q4: How does temperature affect respiration rate in goldfish?
The hypothesis predicts a significant correlation between water temperature and goldfish respiration rate. Since fishes are ectotherms, their metabolic rate depends on environmental temperature. Higher temperatures typically increase dissolved oxygen availability and metabolic activity, potentially increasing gill beat frequency and respiration rate.
Q5: What precautions should be taken when handling fish during respiration experiments?
Gently catch the fish using a net and pinch it carefully without touching the fish itself. Move from lowest to highest temperature to avoid stressing the fish between trials. After completing measurements, slowly mix water from the respiration chamber with tank water over approximately 10 minutes before returning the fish to its home tank.
Q6: How do you properly use a stethoscope to measure heart sounds?
Clean the earpieces with an alcohol swab before inserting them into your ears. Tap the flat metal diaphragm gently to verify the stethoscope is on; if you hear sound, it is active. If not, turn the metal disc and tap again until sound is heard. Place the diaphragm on the skin to listen for heart sounds and Korotkoff sounds during blood pressure measurement.
Q7: What data should be recorded when measuring circulatory function in humans?
Record the subject's name, systolic blood pressure, diastolic blood pressure, and heart rate for each trial in Table 1. Measurements must be taken with the subject both lying down and sitting upright. Both partners should complete measurements in both positions, with partners switching roles to ensure each person serves as both subject and measurer.
