Phase I biotransformation or functionalization entails the chemical transformation of drugs into metabolites via oxidative, reductive, and or hydrolytic reactions.
These reactions introduce or expose polar functional groups, enhancing the drug's hydrophilicity. Their primary purpose is drug detoxification and to facilitate their excretion from the body.
Oxidative reactions in phase I involve oxidation of various carbon systems, including aromatic, olefinic, benzylic, allylic, aliphatic, alicyclic, and carbon-heteroatom systems.
Microsomal enzymes catalyzing these reactions utilize molecular oxygen and NADPH. a reducing agent. So, these enzymes are called mixed-function oxidases.
Examples of drugs undergoing phase I oxidative reactions include phenytoin, diazepam, and codeine.
The resultant phase I metabolites can exhibit decreased, equal, or increased pharmacological activity and serve as substrates for phase II reactions.
