The drug disposition in the body is a complex process and can be studied using two major approaches: the model and the model-independent approaches.
The model approach uses pharmacokinetic models to describe changes in drug concentration over time.
Pharmacokinetic models use a mathematical approach to characterize the drug behavior in patients, predict the drug concentration in the body fluids, calculate the optimum dosage regimens, and evaluate the risk of toxicity.
However, it is important to ensure that the model fits the experimental data accurately.
There are three types of model approaches: compartment, physiological, and distributed parameter models.
The model-independent method, or the noncompartmental analysis, assumes linear kinetics for drugs and can be applied to any compartment model.
This approach involves collecting experimental data after a single dose of the drug.
Non compartmental analysis helps estimate bioavailability, clearance, volume of distribution, and half-life of the drug.
