Abstract

This video demonstrates the preparation and inoculation of anaerobic cultures using oxygen-free sludge under strictly controlled, deoxygenated conditions.

Protocol

Inoculation of the medium

  1. Prepare inoculum from the anaerobic digester.
    1. Add 400 mL of distilled water to a flask and bring it to a boil.
    2. Cool it down (< 30 °C) while permanently flushing the headspace with N2.
    3. Add approximately 100 g of sludge derived from an anaerobic digester.
NOTE: Avoid excessive contact of sludge with oxygen.
    4. Record the exact mass of added sludge for the exact determination of dilution.
    5. Exchange the flask’s headspace with N2 and close it with a butyl rubber septum.
    6. Shake the flask for 30 min at 120 rpm.
