This video demonstrates intranasal delivery of murine herpesvirus in mice to induce a lung-specific immune response. Lung tissue is subsequently processed to obtain cytokine-containing supernatants for downstream assessment of host immune responses.

Protocol

All procedures involving animal models have been reviewed by the local institutional animal care committee and the JoVE veterinary review board.

1. Cytokine Secretion by ELISA in γHV68-infected Mice

  1. Anesthetize gender-matched, 6-8 week-old C57BL/6 (B6) littermates (8-12 mice/group) via intraperitoneally injecting 1.5 mg of ketamine/0.12 mg of xylazine, and inoculate intranasally with 40 PFU(Plaque-forming unit) of γHV68 in 40 µl. At different time points post-infection, euthanize the mice by using CO2 inhalation followed by cervical dislocation.
  2. Open the chest with a left lateral cut along the sternum and cut through th....
