Dr. Roberto Giambruno is a researcher of the Institute for Biomedical Technologies (ITB) of the National Research Council of Italy (CNR) and of the Center for Genomic Science –CGS@SEMM of the Italian Institute of Technology (IIT). His research interest focuses on protein-protein interactions, protein-RNA interactions, post-transcriptional, and post-translational modifications. His research interests also include long noncoding RNA, mass spectrometry-based affinity purification approaches, and nanopore sequencing. He obtained his bachelor’s degree in Biological Sciences from the University of Palermo in 2005. Then, he moved to Rome at the University “La Sapienza” where he completed his master’s degree in Genetic and Molecular Biology in 2008. At the end of 2008, he started as a PhD student in the laboratory of Dr. Giulio Superti-Furga at the Center of Molecular Medicine (CeMM) of the Austrian Academy of Science in Vienna where he was enrolled in the Molecular Signal Transduction Programme at the Medical University of Vienna. In 2014, he completed his PhD studies and moved back to Italy where he joined the laboratory of Davide Gabellini at the San Raffaele Scientific Institute within the San Raffaele International Postdoctoral Programme – Marie Curie Actions. In 2017, he won a 3-year scientist position within the Italian PhDiTalents national programme at the European Institute of Oncology - IEO in the laboratory of Tiziana Bonaldi. From April 2020 to January 2022, he joined as a postdoctoral fellow at the laboratory of Dr. Francesco Nicassio at the CGS@SEMM of IIT in Milan. Starting from February 2022 he started his career as a researcher at the ITB of CNR.

Collection Overview

RNA binding proteins are dynamically interacting with cellular RNA transcripts to regulate their stability, degradation rate, cellular localization, and interaction propensity. Protein-RNA interactions are at the basis of key biological processes such as transcription, RNA splicing, RNA nuclear export, and translation. Specific methods have been developed to study protein-RNA interactions in vitro, in cell, and in vivo models. Especially in the last few years, a particular emphasis has been given to the development of specific methodologies that allows the identification of RNA-protein interactions through OMICS-technologies. In particular, protein-centric and RNA-centric strategies have been coupled to next-generation sequencing and mass spectrometry-based approaches, respectively. This methodology has enhanced the detection of transient interactions as well as the identification of low abundant interactors. In addition, it has been possible to identify and characterize the role that posttranscriptional and posttranslational modifications are playing within RNA and protein interactions.

The aim of this Methods Collection is to include the current methods used to identify, quantify, and characterize RNA-protein interactions occurring both in prokaryotic and eukaryotic cells. Specifically, methods that can determine the dynamics of the interaction of a specific protein and an RNA, such as ITC, BLI, switchSENSE, and the fluorescence-based methods (FRET, FRAP); the structural characteristics of protein-RNA interactions as assessed by NMR, CD, DLS; the global and unbiased identification of RNA-proteins interactions obtained by the adoption of antisense oligos used as probes (RNA-pulldowns, ChIRP, RAP, iDRIP), RNA immunoprecipitation strategies (RIP, CLIP), proximity-ligation techniques (APEX2, RaPID, CaRPID, TRIBE) and organic: aqueous phase separation methods (XRNAX, OOPS, PTEx).
