Harvard Medical School

Dr. Florez is a postdoctoral fellow at Harvard Medical School's Department of Systems Biology. He holds a PhD in Cancer Biology from Heidelberg University and the German Cancer Research Center. His PhD revealed a link between the oncogene MYCN—found in aggressive tumors—and heightened ferroptosis sensitivity, offering a novel approach for targeted cancer treatment. During his Harvard postdoc in the Molecular and Cellular Biology department, he pioneered a CRISPRi tool that proficiently silences genes in bacteria with minimal noise, no leakiness, and reversibility through rapid protein degradation. He authored "Ferroptosis: Mechanism and Diseases," and his ferroptosis research received recognition in New Scientist. His current focus centers on a microfluidic platform using microscopic sorting with single RNA sequencing to detect cancer persister cells post-ferroptosis induction. Dr. Florez aims to synergize his expertise in cancer biology, synthetic biology, and platform development to revolutionize the panorama of cancer therapy.

Collection Overview

The study of cell death has spanned many years of scientific inquiry. Cell death is a vital biological process that helps maintain balance in cellular organisms. It plays a role in tissue renewal, immune response shaping, and embryonic development. Recent research has revealed new cell death pathways, notably ferroptosis—a unique form of iron-mediated cell death. Unlike other forms of cell death, ferroptosis operates independently of known regulatory pathways and is closely tied to cellular metabolism and redox balance. This discovery has filled gaps in our understanding of mammalian metabolism and has implications for drug discovery in areas like cancer, neurodegenerative diseases, and infectious diseases.

This Methods Collection aims to introduce scientists to ferroptosis and offer methods for its detection and analysis, spanning from metabolic analysis, phenotype identification and quantification, and assessing ferroptosis biomarkers in cells or tissues. By presenting a range of techniques, we equip researchers to explore ferroptosis in various experimental contexts. We aim to enhance our understanding of this novel cell death pathway and catalyze innovative work in this scientific field.
