Understanding the mechanisms underlying insect taste reception and perception is critical for learning how insects interact with their environment, locate food, and select mates. Taste receptors are typically located on different body parts of insects, including antennae, mouth parts, tarsae, ovipositors, and wings.

This JoVE Methods Collection sheds light on some primary methods used to study insect taste, including behavioral, physiological, and molecular approaches. Behavioral assays, including the proboscis extension reflex (PER) observed in bees, wasps, and flies, along with feeding preference tests, provide insights into taste-driven behaviors. These assays are complemented by electrophysiological techniques like single sensillum recordings (SSR) and electroantennography (EAG), which measure neuronal responses to gustatory stimuli.

Advances in genetic tools like CRISPR-Cas9 and cloning into heterologous expression systems have enabled precise manipulation of taste receptor genes, allowing researchers to dissect the roles of specific receptors and neurons in taste perception. Calcium imaging and optogenetics enhance our understanding by visualizing and controlling neuronal activity in real time. Molecular computational tools like RNA sequencing, single-cell RNA sequencing, and protein prediction models can be more broadly applied to non-model organisms to facilitate the identification of taste receptors and signaling pathways, as well as predict receptor-ligand interactions to simulate gustatory processing.

This collection presents standardized state-of-the-art experimental procedures that help uncover how insects distinguish between different tastants and the implications of these mechanisms for insect behavior and ecology. Developing sophisticated techniques promises to advance our understanding of insect taste systems, with potential applications in pest control and pollinator management.
