Passive Avoidance Paradigm

The passive avoidance paradigm is a behavioral assay that measures aversive learning and memory by testing an animal’s reluctance to re-enter an environment associated with an unpleasant event. Typically, an animal moves from a brightly lit compartment into a dark compartment, where it receives a mild foot shock; during a later trial, increased latency to enter the dark compartment indicates retention of the learned association. Researchers use this paradigm to assess memory formation, consolidation, and retrieval, and to examine how genetic factors, brain regions, drugs, disease models, or environmental conditions influence behavior.

This work describes a simple behavioral paradigm that allows the analysis of aversive associative learning in adult fruit flies. The method is based on suppressing the innate negative geotaxis behavior due to the association formed between a specific environmental context and an electric shock.

Avoidance is central to chronic pain disability, yet adequate paradigms for examining pain-related avoidance are lacking. Therefore, we developed a paradigm that allows investigating how pain-related avoidance behavior is learned (acquisition), spreads to other stimuli (generalization), can be mitigated (extinction), and how it may subsequently re-emerge (spontaneous recovery).

Ammonia emissions are a major threat to the environment by eutrophication, soil acidification and fine particle formation and stem mainly from agricultural sources.
