Flavin Mononucleotide

Flavin mononucleotide (FMN) is a phosphorylated form of riboflavin that serves as a redox-active cofactor in many enzymes and light-responsive proteins. Its isoalloxazine ring accepts and donates electrons, enabling reversible one- or two-electron reactions as FMN-containing flavoproteins catalyze oxidation-reduction processes. In bioengineering, FMN supports the design of engineered biocatalysts, metabolic pathways, biosensors, and flavin-dependent photoreceptors, including systems that couple light detection to cellular signaling. Understanding FMN binding, electron transfer, and cofactor regeneration helps researchers improve enzyme activity, construct responsive biomaterials, and develop sustainable platforms for chemical synthesis and biotechnology.

Monitoring the Reductive and Oxidative Half-Reactions of a Flavin-Dependent Monooxygenase using Stopped-Flow Spectrophotometry

We describe the use of a stopped-flow instrument to investigate both the reductive and oxidative half-reactions of Aspergillus fumigatus siderophore A (SidA), a flavin-dependent monooxygenase. We then show the spectra corresponding to the species in the reaction of SidA and we calculate the rate constants for their formation.

Mitochondria contain several flavin-dependent enzymes that can produce reactive oxygen species (ROS). Monitoring ROS release from individual sites in mitochondria is challenging due to unwanted side reactions. We present an easy, inexpensive method for direct assessment of native rates for ROS release using purified flavoenzymes and microplate fluorometry.

We demonstrate a simple method for placing cells at desired locations on a substrate. This method patterns cells by flipping a silicone chip containing microwells filled with cells onto the substrate. This method provides a new way to modulate diffusible and juxtacrine signaling between cells.
