Stereo Imaging

Stereo imaging is a technique for reconstructing three-dimensional structure from two or more images captured from different viewpoints, making depth and spatial relationships measurable in bioengineering. The system identifies corresponding features across the images and calculates their disparity, using camera geometry and calibration to triangulate the position of points in space. This approach supports quantitative analysis of biological structures, cell and tissue morphology, movement, and engineered biomaterials without relying on a single two-dimensional view. Stereo imaging can therefore improve spatial characterization in biomechanics, medical imaging, tissue engineering, and the evaluation of complex biological models.

This protocol uses multi-view stereo to generate three-dimensional (3D) models out of uncalibrated sequences of photographs, making it affordable and adjustable to a surgical setting. Strain maps between the 3D models are quantified with spline-based isogeometric kinematics, which facilitate representation of smooth surfaces over coarse meshes sharing the same parameterization.

We describe a new method for counting fishes, and estimating relative abundance (MaxN) and fish density using rotating stereo-video camera systems. We also demonstrate how to use distance from camera (Z distance) to estimate species-specific detectability.
