Tissue Engineering Molds

Tissue engineering molds are three-dimensional templates used to shape biomaterials, cell-laden hydrogels, or developing tissue constructs into defined architectures. By confining a material within a cavity or patterned framework, molds control construct geometry, size, and internal organization while the material cures, gels, or solidifies. Researchers use techniques such as casting and replica molding to create scaffolds that resemble features of native tissues, including pores, channels, and layered structures. These molds support the fabrication of reproducible models for studying cell behavior, testing biomaterials, and developing engineered tissues for regenerative medicine.

Custom Engineered Tissue Culture Molds from Laser-etched Masters

Herein we present a rapid, facile, and low-cost method for fabricating custom polydimethylsiloxane molds that can be used for producing hydrogel-based engineered tissues with complex geometries. We additionally describe results from mechanical and histological assessments conducted on engineered cardiac tissues produced using this technique.

Here, we demonstrate fabrication of collagen-based, tissue constructs containing skeletal myoblasts. These 3-D engineered constructs may be used to replace or repair tissues in vivo. For our purposes, we have designed these as an atrioventricular electrical conduit for the repair of complete heart block[1].

We describe the isolation of neonatal cardiomyocytes and the preparation of the cells for encapsulation in fibrin hydrogel constructs for tissue engineering. We describe methods for analyzing the tissue engineered myocardium after the culture period including active force generated upon electrical stimulation and cell viability and immunohistological staining.
