Gfp Fused Proteins

GFP-fused proteins are engineered proteins in which green fluorescent protein (GFP) is genetically linked to a protein of interest, allowing researchers to visualize its location and behavior in living cells. The fusion gene is expressed to produce a single polypeptide, and GFP forms an internal chromophore that fluoresces when exposed to specific wavelengths of light, provided the fusion retains functional folding. In biological techniques, these constructs help track protein localization, transport, expression, and interactions through fluorescence microscopy and related assays. GFP tagging supports studies of cell organization, signaling, and protein dynamics, while careful design is needed to minimize effects on the target protein’s activity.

A method to monitor ubiquitin-proteasome activity in living cells is described. A degron-destabilized GFP- (GFP-dgn) and a stable GFP-dgnFS fusion protein are generated and transduced into the cell using a lentiviral expression vector. This technique allows to generate a stable GFP-dgn/GFP-dgnFS expressing cell line in which ubiquitin-proteasome activity can be easily assessed using epifluorescence or flow cytometry.
