Alkaline Solutions

Alkaline solutions are aqueous mixtures with a pH above 7, produced when basic substances increase the concentration of hydroxide ions or accept protons. Their alkalinity arises from acid–base reactions in which bases dissociate in water or react with hydrogen ions, shifting chemical equilibria and influencing pH. In biology, alkaline solutions help maintain or adjust conditions for enzyme activity, cellular processes, nucleic acid handling, and laboratory assays. Buffers can moderate changes in alkalinity, making these solutions valuable for studying biochemical reactions and understanding how pH affects molecular structure, physiological function, and experimental outcomes.

The alkaline comet assay measures DNA strand breaks in eukaryotic cells. By adding an Endonuclease III or human 8-oxoguanine-DNA-N-glycosylase digestion step, the assay can efficiently detect oxidative DNA damage. We describe methods for using these assays to detect DNA damage in rat liver.
