Arthrobacter

Arthrobacter is a genus of Gram-positive, high-GC bacteria found primarily in soil and other nutrient-limited environments, where its metabolic flexibility supports ecosystem function and biotechnology. These aerobic actinobacteria undergo a characteristic rod-to-coccus transition as cultures age or environmental conditions change, while their diverse enzymes enable the breakdown of complex organic compounds. Arthrobacter species are therefore studied in microbial ecology, biodegradation, and bioremediation, including the transformation of petroleum-related compounds, pesticides, and other pollutants. Their tolerance of desiccation, cold, and nutrient scarcity also makes them useful models for understanding bacterial survival and for developing strategies to restore contaminated environments.

Source: Cross, T., et al. An Optimized Enrichment Technique for the Isolation of Arthrobacter Bacteriophage Species from Soil Sample Isolates. J. Vis. Exp. (2015).This video demonstrates the isolation of Arthrobacter bacteriophages from a soil sample using enrichment and plaque assay techniques. It highlights the infection, amplification, and visualization of phages as clear lysis zones formed on a bacterial lawn.

Source: Cross, T., et al. An Optimized Enrichment Technique for the Isolation of Arthrobacter Bacteriophage Species from Soil Sample Isolates. J. Vis. Exp.(2015)This video demonstrates the isolation of bacteriophages from a soil sample. A soil sample is collected and suspended in a phage buffer and allowed to settle the sediment. The extract is filtered, and nutrients are added. A vacuum filtration is done to remove microbial contaminants. The filtrate with phages is aliquoted into flasks, and...

Source: Cross, T., et al., An Optimized Enrichment Technique for the Isolation of Arthrobacter Bacteriophage Species from Soil Sample Isolates. J. Vis. Exp.(2015)This video demonstrates the preparation of a bacterial culture for bacteriophage production. It covers the steps of inoculating a soil bacterium into a baffled flask, promoting uniform growth through aeration and shaking, and monitoring cell density using optical density measurements. Once the culture reaches the optimal growth phase,...

2023

Source: Laboratories of Dr. Ian Pepper and Dr. Charles Gerba - The University of Arizona Demonstrating Author: Luisa Ikner Aseptic technique is a fundamental skill widely practiced in the field of environmental microbiology that requires a balance of mindfulness and practice in the laboratory. Proper use of this technique reduces the likelihood of bacterial or fungal contamination of reagents, culture media, and environmental samples. Aseptic technique is also vital to ensure data integrity and...
