Bacterial Burden Quantification

Bacterial burden quantification is the measurement of bacterial abundance in a sample, tissue, or biological system, providing a basis for assessing colonization, infection, and treatment response. Depending on the research question, investigators estimate viable cells by serially diluting samples and counting colony-forming units on agar, or measure total bacterial DNA with molecular assays; optical density can also indicate population growth in liquid culture. These approaches support antimicrobial testing, host-pathogen studies, environmental monitoring, and microbiome research. Accurate quantification helps compare experimental conditions, track changes over time, and evaluate whether an intervention reduces bacterial populations.

Swarming motility is influenced by physical and environmental factors. We describe a two-phase protocol and guidelines to circumvent the challenges commonly associated with swarm assay preparation and data collection. A macroscopic imaging technique is employed to obtain detailed information on swarm behavior that is not provided by current analysis techniques.

Histological Quantification to Determine Lung Fungal Burden in Experimental Aspergillosis

Here we describe a protocol to determine pulmonary fungal burden in mice with invasive aspergillosis by quantification of Gomori's modified methanamine silver staining in histological sections. Use of this method resulted in comparable results with less animals compared to assessment of fungal burden by quantitative PCR of lung fungal DNA.
