Cell To Cell Movement

Cell-to-cell movement is the directed migration of cells through tissues or across surfaces, a fundamental biological process that shapes development, immune responses, wound healing, and disease progression. Cells move by reorganizing the actin cytoskeleton, extending membrane protrusions, forming and releasing adhesion contacts, and generating traction against their surroundings; chemical gradients can guide this movement through chemotaxis. Studying these coordinated behaviors helps explain how cells navigate complex tissue environments, interact with neighboring cells, and cross biological barriers. In biology and biomedical research, cell movement assays support investigations of embryonic development, inflammation, cancer invasion, and tissue repair.

Nanopodia - Thin, Fragile Membrane Projections with Roles in Cell Movement and Intercellular Interactions

Nanopodia are thin but fragile membrane channels that extend up to 100 μm from a cell's leading front or trailing rear and sense the cellular environment. Direct fixation at 37 °C, gentle washing, and avoidance of organic solvents like ethanol, methanol, or acetone and of higher Triton X-100 concentrations are required to observe these cellular structures.

Herein we describe a procedure to capture live images of Drosophila gastrulation. This has enabled us to better understand the apical constriction involved in early development and further analyze mechanisms governing cellular movements during tissue structure modification.

We present a protocol for isolation and culture of primary mouse embryonic palatal mesenchymal cells for time-lapse imaging of two-dimensional (2D) growth and wound-repair assays. We also provide the methodology for analysis of the time-lapse imaging data to determine cell-stream formation and directional motility.
