Centromere Mapping Technique

Centromere mapping techniques identify the position, structure, and genomic boundaries of centromeres, chromosome regions essential for accurate chromosome segregation during cell division. They typically combine physical or sequence-based markers with cytogenetic signals, such as centromere-specific DNA probes or centromere-associated chromatin, to locate these regions and relate them to neighboring genomic sequences. In biology, these maps help characterize repetitive DNA, chromosome organization, and variation among species or individuals. They also support studies of genome assembly, chromosome stability, inheritance, and diseases linked to segregation errors, while improving the interpretation of complex or poorly annotated genomic regions.

Histone Variants at the Centromere

2020

Histone variants are the histone proteins with structural and sequence variations. These variants may be regarded as “mutant” forms that replace their canonical histone counterparts in the nucleosomes. Specific post-translational modifications on the histone variants enable further chromatin complexity and regulate tissue-specific gene expression. The most common histone variants are from histone H2A, H2B, and linker histone H1 families. However, several variants of histone H3 variants are also...

Here we report protocols to detect endogenous and exogenous centromere-kinetochore proteins in human cells and quantify these protein levels at centromeres-kinetochores by indirect immunofluorescent staining through the use of fixation (paraformaldehyde, acetone, or methanol fixation).

Live Imaging Characterization of Centromere Movements During Male Meiotic Prophase in Arabidopsis thaliana

2025

Chromosome movement during meiotic prophase is a poorly understood phenomenon that is fundamental to spore formation. This protocol describes a method to acquire, analyze, and quantify centromere movements during meiotic prophase in Arabidopsis thaliana male meiocytes.

Fate Mapping

2023

Fate mapping is a technique used to understand how embryonic cells divide, differentiate, and migrate during development. In classic fate mapping experiments, cells in different areas of an embryo are labeled with a chemical dye and then tracked to determine which tissues or structures they form. Technological improvements now allow for individual cells to be marked and traced throughout embryonic development and adulthood. This video reviews the concepts behind fate mapping, and then details a...

Motor Maps

Source: Laboratories of Jonas T. Kaplan and Sarah I. Gimbel—University of Southern California One principle of brain organization is the topographic mapping of information. Especially in sensory and motor cortices, adjacent regions of the brain tend to represent information from adjacent parts of the body, resulting in maps of the body expressed on the surface of the brain. The primary sensory and motor maps in the brain surround a prominent sulcus known as the central sulcus. The cortex...
