Cerebellar White Matter

Cerebellar white matter is the inner, myelinated axon network of the cerebellum that connects its cortical regions with deep cerebellar nuclei and other brain structures. Its branching pattern, known as the arbor vitae, enables rapid transmission of signals because myelin electrically insulates axons and increases conduction efficiency. These pathways carry information involved in coordinating movement, maintaining balance, regulating muscle tone, and supporting aspects of motor learning. Studying cerebellar white matter through anatomy, histology, neuroimaging, and developmental biology helps researchers understand normal neural organization and identify structural changes associated with neurological disorders.

Here we present methodology for the production of a focal stroke in murine white matter by local injection of an irreversible endothelial nitric oxide synthase (eNOS) inhibitor (L-Nio). Presented are two stereotactic variations, retrograde neuronal tracing, and fresh tissue labeling and dissection that expand the potential applications of this technique.

This video demonstrates the precise technique for inducing a white matter stroke in an anesthetized mouse by injecting a nitric oxide inhibitor at targeted brain coordinates within the white matter. This leads to blood vessel constriction and inhibition of blood supply, resulting in neuronal damage and a brain stroke.

DTI of the Visual Pathway - White Matter Tracts and Cerebral Lesions

Diffusion tensor imaging (DTI) was performed to try to depict the major parts of the visual pathway. The goal was to use an FDA approved standard commercial workstation which could be used for everyday routine in order to try to reduce postoperative damage of the visual pathway in the patients.
