Cover Slip Sealing

Cover slip sealing is a microscopy technique that secures a cover slip over a mounted biological specimen to preserve the preparation and improve imaging stability. A sealant, such as mounting medium or a suitable edge seal, is applied around the cover slip, creating a barrier that limits evaporation, fluid movement, and entry of contaminants while maintaining the specimen’s position. In biology, sealed preparations support consistent observation of cells, tissues, and microorganisms, particularly during repeated imaging or storage. The method can also help protect fluorescent or stained samples, improving reproducibility in light microscopy and preserving valuable slides for later analysis.
