Dna Template

A DNA template is a strand of DNA that provides the sequence information needed to build a complementary nucleic acid strand, making it central to genetic information transfer. During DNA replication, DNA polymerase reads the template strand and adds complementary nucleotides, while RNA polymerase uses a DNA template to synthesize messenger RNA during transcription. In biology, DNA templates support gene expression, inheritance, and laboratory methods such as polymerase chain reaction, DNA sequencing, and cloning. Understanding template-directed synthesis helps explain how cells accurately copy and use genetic information, while also enabling researchers to analyze genes and develop molecular diagnostics.

The combination of chromatin immunoprecipitation and ultra-high-throughput sequencing (ChIP-seq) can identify and map protein-DNA interactions in a given tissue or cell line. Outlined is how to generate a high quality ChIP template for subsequent sequencing, using experience with the transcription factor TCF7L2 as an example.
