Ero1 Oxidoreductase

Ero1 oxidoreductase is an endoplasmic reticulum flavoprotein that maintains oxidative protein-folding conditions by promoting disulfide bond formation in newly synthesized proteins. It reoxidizes protein disulfide isomerase (PDI), transferring electrons from reduced PDI to molecular oxygen and generating hydrogen peroxide as a byproduct; oxidized PDI can then introduce and rearrange disulfide bonds in substrate proteins. In biology, Ero1 activity links protein maturation with endoplasmic reticulum redox homeostasis and must be regulated to balance efficient folding against oxidative stress. Studying Ero1 helps clarify secretory-pathway function, the unfolded protein response, and cellular mechanisms that respond to protein-folding disturbances.

2025

Source: Alluri, H., et.al. Oxygen-Glucose Deprivation and Reoxygenation as an In Vitro Ischemia-Reperfusion Injury Model for Studying Blood-Brain Barrier Dysfunction. J. Vis. Exp. (2015).The video demonstrates the impact of oxygen-glucose deprivation and subsequent reoxygenation on rat brain microvascular endothelial cells (RBMECs) by simulating hypoxic and nutrient-deprived conditions. These conditions result in ATP depletion, increased intracellular calcium, and the generation of reactive...

An In Vitro Assay to Evaluate the Effect of Antibodies on Candida tropicalis Biofilm

2025

This video demonstrates an in vitro technique to study the effect of antibodies on fungal biofilms. The pathogenic fungus Candida tropicalis secretes a virulence factor secreted aspartyl proteinase 2 (Sap2), facilitating biofilm formation. Upon introducing a heat-inactivated serum containing Sap2-specific antibodies that inhibit biofilm maturation, the viability of the fungal cells is assessed using a chromogenic assay.

Methyl Thiazolyl Tetrazolium or MTT Assay: A Colorimetric Assay to Measure Drug Resistance via Determination of Metabolic Activity in Cancer Cells

2023

This video describes the colorimetric assay, methyl thiazole tetrazolium (MTT) assay, to measure drug-induced cytotoxicity in cancer cells and determine their metabolic activity. This assay is based on the metabolic conversion of tetrazolium salts into colored products, reflecting the mitochondrial activity of cells.

An Assay to Evaluate Drug Efficacy against a Mycobacterium tuberculosis Infection Model

2025

This video demonstrates an assay to evaluate the efficacy of drugs against Mycobacterium tuberculosis (Mtb) infection. An in vitro infection model containing Mtb/macrophage aggregates is generated that mimics in vivo infection. Upon introducing suitable antibiotics, their efficacy is tested via resuzarin assay to monitor the survival of bacteria with increasing drug concentrations.
