Fixative Instillation

Fixative instillation is a preservation technique that introduces a chemical fixative into a biological specimen, organ, or tissue compartment to maintain its structure for examination. The solution penetrates the target region and stabilizes cellular components, commonly by cross-linking proteins or precipitating macromolecules, while limiting enzymatic degradation and microbial decay. In biology, instillation supports histological processing, anatomical studies, and microscopy when uniform preservation of internal surfaces or hollow structures is important. By retaining tissue architecture and cellular detail, the method improves the reliability of morphological observations and downstream analyses.

Intratracheal instillations deliver solutes directly into the lungs. This procedure targets the delivery of the instillate into the distal regions of the lung, and is therefore often incorporated in studies aimed at studying alveoli. We provide a detailed survival protocol for performing intratracheal instillations in mice.
