Fly Transfer Techniques

Fly transfer techniques are laboratory methods for safely moving flies between culture containers while preserving their viability and minimizing contamination. In Drosophila research, flies are typically immobilized with carbon dioxide or cold anesthesia, then guided with a soft brush, aspirator, or transfer vial into fresh food medium; careful handling limits injury, escape, and unintended changes to the culture. These procedures support routine stock maintenance, controlled mating, genetic crosses, developmental studies, and behavioral experiments. Consistent transfer practices help researchers maintain defined genotypes, synchronize populations, and obtain reliable results across biological replicates.

This video describes one method to build a fly aspirator, which can be used to transfer insects between enclosures without anesthesia. The aspirator in the example protocol was prepared for an experiment to quantify individual grooming...

A Septic Pinprick Technique to Introduce Systemic Bacterial Infection in Flies

2025

This video demonstrates a technique to infect Drosophila by introducing a pathogenic bacteria directly into their body cavity. This method bypasses epithelial barriers and other passive defense mechanisms, allowing for the induction of systemic infection. Upon infection, the resulting immune response is assessed by monitoring fly mortality and performing downstream assays.

Drosophila has emerged as a significant model system for dissecting the cellular and molecular underpinnings of behavioral responses to alcohol. Here we present a protocol for the collection of alcohol sensitivity data in a circadian context that can be easily applied to other experiments and is well-suited for undergraduate research.
