Ligation-mediated Primer Extension

Ligation-mediated primer extension is a molecular biology method that combines sequence-specific primer extension with ligation to identify, copy, or analyze nucleic acid targets. A primer first hybridizes to a complementary DNA or RNA template, and a polymerase adds nucleotides from the primer’s 3′ end; the resulting extension product is then joined to an adjacent nucleic acid or adapter through ligase activity, creating a defined molecule for downstream detection. This approach can improve sequence specificity and is useful for mutation analysis, transcript characterization, targeted sequencing, and other assays requiring precise recognition of a target sequence.

We present a method of targeted ancient DNA sequence retrieval, which we used to reconstruct the complete mitochondrial genomes of five Neandertal individuals. Comparison of these sequences with present day humans suggests that Neandertals had a long term low effective population size.

Primer-Free Aptamer Selection Using A Random DNA Library

SELEX protocols comprise multiple rounds of selection, each of which require regeneration of bound ligands, which in turn require fixed primer sequences flanking the random library regions. These fixed primer sequences can interfere with the selection process (false positives and negatives). Here we present a primer-free protocol.
