Maize Seedling Injection

Maize seedling injection is a laboratory technique that delivers a measured solution directly into the tissues of young maize plants, enabling controlled study of seedling biology. The method uses a fine needle or similar implement to penetrate a selected tissue and deposit the material at a defined location and volume, while careful handling helps limit physical damage. Researchers can use this approach to introduce experimental treatments, test tissue responses, and assess how seedlings react to localized biological or chemical inputs. Its controlled delivery supports reproducible experiments in plant physiology, developmental biology, and maize research.

An Agrobacterium-based injection (agroinjection) protocol is presented for the inoculation of foxtail mosaic virus and sugarcane mosaic virusclones into maize seedlings. Inoculation in this manner leads to viral infection, virus-induced gene silencing of marker genes, and viral overexpression of GFP.

Three-Dimensional Immunolocalization In Maize Meiocytes

2026

This immunolocalization protocol in male maize meiocytes maintains the three-dimensional integrity of the cells during meiosis. It allows the dynamic tracking of the intracellular localization of up to three independent antibodies in preserved paraformaldehyde-fixed material, in addition to DAPI, which stains DNA.

Source: Hassan, J. A., et al. High-Throughput Identification of Resistance to Pseudomonas syringae pv. Tomato in Tomato using Seedling Flood Assay. J. Vis. Exp. (2020).This video demonstrates a method to screen for bacterial resistance in different tomato strains using ten-day-old seedlings inoculated with a bacterial suspension. It explains how bacteria invade through stomata, deliver effector proteins to suppress plant immunity, and how plant responses differ between susceptible and resistant...
