Nod Scid Mice

NOD scid mice are laboratory mouse models with severe combined immunodeficiency, making them valuable for studying immune function and human cell behavior in vivo. The scid mutation disrupts V(D)J recombination, preventing development of mature T and B lymphocytes, while the nonobese diabetic (NOD) background contributes defects in innate immune responses, including reduced natural killer-cell activity and complement function. Because they accept many transplanted human or mouse cells with limited immune rejection, researchers use these mice for xenografts, tumor biology, infectious-disease studies, stem-cell research, and preclinical evaluation of therapies. Their engraftment capacity also supports development of humanized immune-system models.

This protocol provides a method to establish humanized mice (hu-NSG) via intrahepatic injection of human hematopoietic stem cells into radiation-conditioned neonatal NSG mice. The hu-NSG mouse is susceptible to HIV infection and combinatorial antiretroviral therapy (cART) and serves as a suitable pathophysiological model for HIV replication and latency investigations.

We have described a protocol for performing partial hepatectomy (PHx) and cell transplantation via spleen in NOD.SCID (NOD.CB17-Prkdcscid/J) mice. In this protocol, an incision is made to expose and resect the left lobe of the liver followed by another incision for the intrasplenic transplantation of cells.

Our protocol was developed to cleanly and easily deliver islets or cells under the kidney capsule of mice. Cells are concentrated into pellets in the final tubing used for transplanting the cells under the kidney capsule. The ease of this technique reduces stress to the cells and the mouse.

This video demonstrates an assay to generate a humanized xenograft mouse model. An immunocompromised mouse is injected with cyclophosphamide to cause immune cell depletion. Patient-derived tumor cells mixed with human peripheral blood mononuclear cells (PBMCs) in a hydrogel are subcutaneously injected into the mouse, forming a tumor with infiltrating T cells.
