Od 600 Measurement

OD600 measurement is a spectrophotometric method for estimating the concentration and growth of microbial cells in a liquid culture by measuring optical density at a wavelength of 600 nanometers. As cells increase in number, they scatter more light, causing the culture’s absorbance or turbidity reading to rise; the result is commonly used as a relative indicator of biomass rather than a direct cell count. In biology, researchers use OD600 to monitor growth curves, compare culture conditions, standardize inocula, and determine when cultures reach a desired growth phase. Reliable measurements require consistent sample preparation, appropriate dilution, and a blank containing the culture medium.

Standardized Method for Measuring Collection Efficiency from Wipe-sampling of Trace Explosives

Optimized sampling protocols and the development of new wipe materials can be facilitated by standardized measurements of collection efficiency from wipe-sampling. Our approach for sampling trace explosives uses an automated device to control speed, force, and distance during wipe-sampling followed by extraction of collected explosives.

Vacuolar and cytosolic pH can be measured in live yeast (S. cerevisiae) cells using ratiometric fluorescent dyes localized to specific cellular compartments. We describe procedures for measuring vacuolar pH with BCECF-AM, which localizes to the vacuole in yeast, and cytosolic pH with a cytosolic ratiometric pH-sensitive GFP (yeast pHluorin).
