Optimal Cutting Temperature

Optimal Cutting Temperature (OCT) is a water-soluble embedding compound used to support biological specimens during cryostat sectioning, enabling thin frozen sections for microscopy and analysis. It surrounds tissue, fills gaps, and freezes into a firm matrix that stabilizes the sample while a chilled blade cuts through it; the matrix can then be removed during aqueous staining procedures. In biology, OCT embedding helps preserve tissue architecture and supports the detection of cellular antigens, enzymes, and nucleic acids, making it useful for histology, immunofluorescence, developmental studies, and research on disease-related tissue changes.

Sphingolipids are bioactive metabolites with well-established roles in human disease. Characterizing alterations in tissues with mass-spectrometry can reveal roles in disease etiology or identify therapeutic targets. However, the OCT-compound used for cryopreservation in biorepositories interferes with mass-spectrometry. We outline methods to analyze sphingolipids in human tissues embedded in OCT with LC-ESI-MS/MS.
