Organoid Drug Testing

Organoid drug testing is a method that evaluates drug effects using three-dimensional, self-organizing cell cultures that model key features of organs or tumors. Researchers grow organoids from stem cells or patient-derived cells, expose them to candidate compounds, and measure outcomes such as viability, growth, morphology, or molecular responses under controlled conditions. This approach can capture aspects of tissue architecture and cell-to-cell interactions that conventional two-dimensional cultures may miss. In biology and biomedical research, organoid drug testing supports disease modeling, toxicity assessment, therapeutic screening, and investigation of patient-specific treatment responses, helping bridge laboratory experiments and clinical decision-making.

This protocol describes an assay for measuring CFTR function and CFTR modulator responses in cultured tissue from subjects with cystic fibrosis (CF). Biopsy-derived intestinal organoids swell in a cAMP-driven fashion, a response that is defective (or strongly reduced) in CF organoids and can be restored by exposure to CFTR modulators.
