Outer Mitochondrial Membrane

The outer mitochondrial membrane is the mitochondrion’s permeable boundary, separating the organelle from the cytosol while coordinating exchange and signaling essential for cellular energy metabolism. Its channel proteins, including voltage-dependent anion channels, permit many small metabolites and ions to pass, whereas specialized translocase complexes import nuclear-encoded proteins into mitochondria; interactions with the inner membrane and cytosolic factors regulate this traffic. During apoptosis, increased permeability can release cytochrome c and activate caspases, making the membrane central to programmed cell death. Studying its composition, dynamics, and protein transport mechanisms helps explain metabolic disorders, neurodegeneration, and potential therapeutic targets in cancer biology.

Mitochondria can utilize the electrochemical potential across their inner membrane (ΔΨm) to sequester calcium (Ca2+), allowing them to shape cytosolic Ca2+ signaling within the cell. We describe a method for simultaneously measuring mitochondria Ca2+ uptake and ΔΨm in live cells using fluorescent dyes and confocal microscopy.
