Pbmc Culture

PBMC culture is the ex vivo maintenance of peripheral blood mononuclear cells, primarily lymphocytes and monocytes, under controlled laboratory conditions to study human immune-cell behavior. Researchers typically isolate these cells from peripheral blood by density-gradient centrifugation, then maintain them in suitable culture medium, using mitogens, antigens, cytokines, or other stimuli to activate defined cellular responses. These cultures support analysis of proliferation, cytokine secretion, cell signaling, cytotoxicity, and immune-cell interactions. In biology and biomedical research, PBMC culture provides a practical model for investigating infection, inflammation, immunotherapy, vaccine responses, and drug effects while retaining diverse immune-cell populations.

A Co-culture Method to Investigate the Crosstalk Between X-ray Irradiated Caco-2 Cells and PBMC

We present a protocol to investigate the crosstalk between X-ray-irradiated Caco-2 and peripheral blood mononuclear cells (PBMC). The protocol starts with Caco-2 irradiation and set-up of the co-culture with PBMC; subsequently, trans-epithelial electrical resistance is measured regularly over 48 h and western blot performed in both Caco-2 and PBMC.

This video demonstrates an assay to generate a humanized xenograft mouse model. An immunocompromised mouse is injected with cyclophosphamide to cause immune cell depletion. Patient-derived tumor cells mixed with human peripheral blood mononuclear cells (PBMCs) in a hydrogel are subcutaneously injected into the mouse, forming a tumor with infiltrating T cells.

Unlike cell-free HIV-1 particles, infected CD4+ T cells are effectively sensed by plasmacytoid dendritic cells (pDCs). This manuscript describes a method where peripheral blood mononuclear cells (PBMCs) or isolated pDCs are co-cultured with HIV-1 infected T cells to evaluate innate sensing by pDCs as assessed by release of type-I IFN.
