Perfusate Composition Control

Perfusate composition control is the deliberate regulation of the chemical and physical properties of a fluid circulated through an isolated organ, tissue, or biological model to maintain physiological conditions outside the body. During perfusion, researchers adjust components such as oxygen, carbon dioxide, nutrients, electrolytes, buffers, and osmotic agents while monitoring pH, temperature, pressure, and flow to preserve cellular metabolism and viability. This control supports ex vivo organ perfusion, tissue engineering, disease modeling, and pharmacological testing, enabling investigators to distinguish biological responses caused by experimental treatments from those produced by inadequate perfusion conditions.

Scaffolds for tissue engineering need to recapitulate the complex biochemical and biophysical microenvironment of the cellular niche. Here, we show the use of interfacial polyelectrolyte complexation fibers as a platform to create composite, multi-component polymeric scaffolds with sustained biochemical release.

The porcine model of liver normothermic machine perfusion (NMP), described here, can be successfully used to study NMP as a preservation strategy, a tool for viability assessment, and a platform for organ repair. It holds a high translational value, however it is technically challenging and labor-intensive.
