Product Inhibition

Product inhibition is a form of enzyme regulation in which a reaction product reduces the activity of the enzyme that generated it. The product may compete with the substrate for the enzyme’s active site or bind elsewhere, changing enzyme conformation and lowering catalytic efficiency; inhibition can therefore depend on product concentration and substrate availability. In biology, product inhibition helps balance metabolic pathways by preventing unnecessary accumulation of downstream compounds. Studying this process supports analysis of enzyme kinetics, pathway control, cellular metabolism, and feedback regulation, while also informing biotechnology and the design of strategies to improve biochemical production.

Bacterial pyomelanin production results in increased resistance to oxidative stress and virulence. We report on techniques that can be used to determine inhibition of pyomelanin production and assay the resulting increase in sensitivity to oxidative stress in bacteria, as well as determine antibiotic minimum inhibitory concentration (MIC).

We describe the enzyme-linked lectin assay (ELLA) for measuring influenza neuraminidase (NA)-inhibition antibody titers in sera. The assay uses peanut agglutinin to quantify galactose residues that become accessible when NA removes sialic acid from fetuin-coated, 96-well plates.

This video demonstrates an assay measuring influenza neuraminidase (NA) inhibiting antibody titers in a serum sample. A serum sample containing antibodies against NA is added to a fetuin-coated microwell plate.
