Radio Labeled Proteins

Radiolabeled proteins are proteins linked to radioactive isotopes so their abundance, movement, interactions, or fate can be measured in biological systems. The isotope may be incorporated during protein synthesis or chemically attached to an amino acid residue; as it decays, emitted radiation is detected by methods such as autoradiography, scintillation counting, or gamma spectroscopy. In biology, these probes help researchers follow protein trafficking, quantify synthesis and degradation, identify binding partners, and measure transport across cells or tissues. With appropriate controls and radiation safety practices, radiolabeled proteins provide sensitive kinetic and localization data that complement fluorescence-based approaches in molecular and cellular research.

This paper describes a method for the measurement of fuel oxidation in Drosophila melanogaster in which trace amounts of specific radiolabeled metabolic substrates are fed to flies. The exhaled radiolabeled CO2 that is a produced from fuel oxidation is collected and measured.

This video demonstrates an in vitro technique for estimating amino acid uptake by bone marrow-derived stromal cells. Na+-dependent amino acid transporters mediate the uptake of radiolabeled amino acids inside the cells, and the cellular uptake is estimated by measuring the radioactivity.
