Serial Centrifugation

Serial centrifugation is a laboratory technique that separates suspended particles through a sequence of centrifugation steps, making it important for isolating cellular components and other biological materials. Each spin applies centrifugal force under defined conditions, causing larger or denser particles to sediment first; the remaining supernatant is then transferred to a new tube and centrifuged at a higher speed to collect smaller components. In biology, this approach supports differential fractionation of cell lysates into enriched fractions containing structures such as nuclei, mitochondria, membranes, or ribosomes. The resulting fractions enable biochemical analysis, microscopy, protein studies, and investigation of cellular organization.
