Small Volume Myography

Small volume myography is an ex vivo technique for measuring the contractile and dilatory behavior of isolated small blood vessels under controlled laboratory conditions. A vessel segment is mounted in a compact chamber and exposed to defined pressures or vasoactive substances while changes in force or lumen diameter are recorded, allowing researchers to assess smooth muscle and endothelial responses. In biology, this method helps characterize vascular reactivity, investigate mechanisms that regulate blood flow, and identify how disease, drugs, or experimental treatments alter microvascular function. Its low sample-volume design supports efficient studies of specialized or limited tissue samples.

In pressure myography, an intact small segment of a vessel is mounted onto two small cannulas and pressurized to a suitable luminal pressure. Here, we describe the method to measure vasorelaxation response of the mouse 3rd order mesenteric arteries in c57 and sGCα1-/- mice using pressure myography.
