Sonication Lysate

Sonication lysate is a biological cell or tissue extract produced by disrupting samples with high-frequency ultrasonic energy, providing access to intracellular proteins, nucleic acids, and other molecules. During sonication, alternating pressure waves generate cavitation bubbles whose collapse creates localized shear forces that rupture cell membranes; controlled pulse duration, amplitude, temperature, and sample volume help limit heat-related damage. The resulting lysate supports protein quantification, enzyme assays, immunoblotting, and downstream purification, while efficient homogenization can improve recovery from difficult samples. Because sonication conditions influence fragment size and molecular integrity, they must be optimized for the organism, sample type, and intended analysis.

Differential diagnostics in painful arthroplasty is crucial for treatment success. Joint aspiration is routinely performed preoperatively. Multiplex polymerase chain reaction of the joint aspirate and the sonication fluid, a feasible tool for quick pathogen detection from these samples is described.
