T Cell Receptor Imaging

T cell receptor imaging is a set of microscopy-based methods used to visualize the location, organization, and movement of T cell receptors during immune recognition. Researchers label receptors or associated signaling proteins with fluorescent probes and track their behavior as T cells contact antigen-presenting cells, where peptide-MHC binding can reorganize receptors at the immunological synapse and initiate signaling. These approaches reveal how receptor clustering, mobility, and interactions with co-receptors contribute to T cell activation. In biology and immunology, T cell receptor imaging supports studies of immune-cell communication, antigen specificity, autoimmune disease, infection, and the development of engineered T cell therapies.

Here we present a protocol to image cells expressing green fluorescent protein-tagged angiotensin type 1a receptors during endocytosis initiated by angiotensin II treatment. This technique includes labeling lysosomes with a second fluorescent marker, and then utilizing software to analyze the co-localization of receptor and lysosome in three dimensions over time.
