Transgenic Mouse Creation

Transgenic mouse creation is a genetic engineering method that produces mice carrying an introduced DNA sequence, allowing researchers to study gene function in a living mammal. Typically, scientists assemble a transgene and introduce it into a fertilized egg or embryonic stem cell, where it can integrate into the genome; cells or embryos carrying the construct are then used to generate offspring and establish a heritable mouse line. These models can reveal how genes influence development, physiology, and disease, while tissue-specific or inducible expression systems help link genetic changes to particular cells or conditions. Transgenic mice also support drug evaluation and the study of potential therapeutic strategies in biology and medicine.

Source: Breid, S., et. al. Bioluminescence Imaging of Neuroinflammation in Transgenic Mice After Peripheral Inoculation of Alpha-Synuclein Fibrils. J. Vis. Exp. (2017)This video demonstrates the bioluminescence imaging of neuroinflammation in transgenic mice, showcasing luciferase-luciferin reactions in activated astrocytes. The protocol enables non-invasive quantification of astrocytic activation, aiding studies on CNS inflammation and therapeutic efficacy.

Imaging Transgenic Beta-Cells in an Immunodeficient Mouse Challenged with Diabetic Splenocytes

This video demonstrates an imaging-based method to visualize transplanted luciferase-expressing beta cells into a non-obese diabetic immunodeficient mouse. The introduction of splenocytes from a diabetic mouse leads to progressive beta cell destruction, visually confirmed by declining luminescence after substrate introduction.

C. elegans is usually grown on solid agar plates or in liquid cultures seeded with E. coli. To prevent bacterial byproducts from confounding toxicological and nutritional studies, we utilized an axenic liquid medium, CeHR, to grow and synchronize a large number of worms for a range of downstream applications.

Creation of Reversible Cholestatic Rat Model

2011

Cholestasis is a clinical condition commonly encountered by both surgeons and gastroenterologists. Creation of a reversible cholestatic rat model can be challenging in view of the smaller size and unique hepatopancreatobiliary anatomy in rats. This video article demonstrates the creation of a reversible cholestatic model.
