Viral Antigen Detection

Viral antigen detection is a diagnostic approach that identifies proteins from a virus in clinical or biological samples, providing evidence of viral presence. In these assays, antibodies selectively bind target antigens, and the resulting immune complexes generate a visible, colorimetric, fluorescent, or instrument-read signal through formats such as lateral-flow tests or enzyme-linked immunosorbent assays. The method supports rapid screening, outbreak response, and monitoring of viral circulation when timely results are important. Because antigen tests detect viral components directly rather than relying on host antibody production, they can complement nucleic acid testing and serology in biology and infectious-disease research.

2025

In this video, we demonstrate the process for detecting antibody isotypes in multiple human serum samples using a microarray slide printed with antigenic subtypes of influenza virus strains.

Suppressing Hepatitis B Virus Replication Using Viral Antigen-Specific T Cells

2025

The video demonstrates an assay using human induced pluripotent stem cell (iPSC)-derived CD8+ T cells to inhibit hepatitis B virus (HBV) replication in mice. These T cells, engineered with HBV-specific receptors, are transferred into transgenic mice. Replication-competent HBV genome plasmids are then injected directly into the liver, enabling viral DNA uptake. Once viral antigens are expressed and presented by hybrid MHC-I molecules, the engineered T cells recognize and bind to these complexes.

An Immunohistochemistry Staining for the Detection of Rabies Virus Antigens

2025

This video demonstrates immunohistochemistry staining for rabies virus antigen detection in infected mouse brain tissue. The process includes deparaffinization and alcohol treatment, followed by proteolytic treatment to enhance antigen accessibility. Immunohistochemistry staining reveals red precipitates, confirming the presence of rabies antigens in the tissue section.

Immunostaining to Visualize Viral Antigen and Host Protein Colocalization in the Insect Gut

Source: Zhang, L., et al. Immunofluorescent Labeling of Plant Virus and Insect Vector Proteins in Hemipteran Guts. J. Vis. Exp. (2021)The video demonstrates the preparation of gut tissue from a plant virus–infected insect vector for fluorescence imaging. Fixed guts were washed, permeabilized, and incubated with two fluorescent antibodies in a blocking solution—one targeting a viral antigen in vesicles, the other a vesicle-associated membrane protein. The tissue was counterstained for actin...

The Hemagglutination Inhibition Assay to Detect Serum Antibodies Against a Target Antigen

2025

The video demonstrates a hemagglutination inhibition assay for detecting serum antibodies against specific antigens. Confirmation of hemagglutination inhibition in the test well and hemagglutination in the control well is achieved through distinct visual patterns. This assay plays a crucial role in assessing immune response, vaccine effectiveness, and studying viral infections in virology and immunology.
