Cell Population Isolation

Cell population isolation is the process of separating defined groups of cells from a mixed sample, enabling their properties and behavior to be studied with greater precision. In cancer research, tissue dissociation releases individual cells, which can then be separated according to surface markers, density, size, adhesion, or other physical properties using methods such as magnetic-activated cell sorting, fluorescence-activated cell sorting, density gradients, or selective culture conditions. Isolating tumor, immune, stromal, or drug-resistant populations supports molecular profiling, functional assays, and analysis of tumor heterogeneity. These approaches improve experimental resolution and help identify cellular mechanisms that influence cancer progression and treatment response.

Side Population Assay: A Method to Isolate Side Population of Tumor Cells from Zebrafish

The following protocol describes a method for isolating the side population of T-cell Acute Lymphoblastic Leukemia (T-ALL) tumor cells, consisting of stem cells, from an adult Zebrafish. The side population lacks prominent cell markers and hence we use Hoechst 33342 dye efflux assay to isolate them.

Here, we describe a technique to isolate the side population cells from a zebrafish model of myc-induced T-cell acute lymphoblastic leukemia (T-ALL). This side population assay is highly sensitive and is described for zebrafish T-ALL, but it may be applicable to other malignant and non-malignant zebrafish cell types.

Human cardiac tissue harbours multipotent perivascular precursor cell populations that may be suitable for myocardial regeneration. The technique described here allows for the simultaneous isolation and purification of two multipotent stromal cell populations associated with native blood vessels, i.e. CD146+CD34- pericytes and CD34+CD146- adventitial cells, from the human myocardium.
