Ecm Gel Handling

ECM gel handling is the controlled preparation, storage, and application of extracellular matrix gels that provide a three-dimensional environment for cells in laboratory studies. These protein-rich materials often remain fluid when chilled but polymerize into supportive networks at physiological temperatures, so researchers must manage temperature, timing, sterility, and gentle pipetting to preserve gel structure. In cancer research, ECM gels support tumor spheroids, organoids, and 3D cell cultures, enabling analysis of cell growth, morphology, invasion, and responses to therapeutic compounds. Consistent handling improves experimental reproducibility and helps model how cancer cells interact with the tissue microenvironment.

Gel Purification

2023

Gel purification is used to recover DNA fragments after electrophoretic separation. DNA recovery from an agarose gel includes three basic steps: binding, washing and eluting from a silica column. DNA is believed to bind to silica in the presence of high salt via a salt bridge. Following binding, DNA is washed of impurities and eluted under low salt conditions disrupting this interaction. This video goes through a step-by-step, generalized procedure for cutting out a band from the gel, gel...

This video describes the preparation of a homogenized and digested extracellular matrix suspension from decellularized and lyophilized tissues. This technique helps in fabricating ECM-derived porous foams and microcarriers for use as substrates in advanced 3D cell culture models.

Handling Chemical Spills

Source: Robert M. Rioux & Taslima Zaman, Pennsylvania State University, University Park, PA Since chemicals are commonly used for laboratory research purposes, it is extremely important to be adequately prepared to handle chemical spills or accidental release of hazardous chemicals, which can happen at any time. No matter how minor a spill may be, the inability to respond in an emergency situation could severely endanger public health or the environment. All chemical spills must be properly...

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Acclimation and Positive Reinforcement Training Pigs are social animals that respond best to calm, low-stress handling techniques. Acclimation and positive reinforcement training are important refinement strategies recommended...
