Human Tissue Preparation

Human tissue preparation is the controlled collection, preservation, processing, and sectioning of human tissue so its cellular architecture and molecular features can be examined reliably. In cancer research, specimens may be fixed to stabilize proteins and structures, embedded in paraffin, cut into thin sections, and stained, while fresh or frozen tissue can preserve nucleic acids for molecular assays; careful handling and standardized conditions limit distortion and degradation. These preparations support histopathology, tumor classification, biomarker evaluation, genomic analysis, and comparisons between malignant and normal tissue. High-quality preparation therefore links patient specimens to reproducible diagnoses, mechanistic studies, and development of targeted therapies.

We present a protocol for ex vivo cultivation of human ventricular myocardial tissue. It allows for detailed analysis of contraction force and kinetics, as well as the application of pre- and afterload to mimic the in vivo physiological environment more closely.

This protocol successfully prepared sufficient high-quality neovascular tissue from glioma samples using laser-capture microdissection for quantitative proteomics analysis of tumor angiogenesis, overcoming the limitations of studies that traditionally focus mainly on angiogenic factors and providing a large-scale proteomics profile of glioma angiogenesis.

Tissue Engineering of a Human 3D in vitro Tumor Test System

Methods to create human 3D tumor tissues as test systems are described. These technologies are based on a decellularized Biological Vascularized Scaffold (BioVaSc), primary human cells and a tumor cell line, which can be cultured under static as well as under dynamic conditions in a flow bioreactor.
