Raji Cells

Raji cells are a human B-lymphoblastoid cell line derived from Burkitt lymphoma and widely used as an experimental model in cancer research. These Epstein-Barr virus-positive cells grow as nonadherent clusters in suspension culture, retaining B-cell markers such as CD20 and proliferating under appropriate nutrient and environmental conditions. Researchers use Raji cells to study lymphoma biology, B-cell signaling, antigen presentation, and interactions between tumor cells and immune cells. Their reproducible growth and defined malignant origin also support drug screening, antibody evaluation, and investigations of immunotherapies, making them a practical model for connecting cellular mechanisms with potential cancer treatments.

Immunological Synapse Formation Between Helper T Cells and Antigen-Presenting Cells

This video demonstrates the in vitro study of immunological synapse formation between an antigen-presenting cell and helper T lymphocytes using a fluorescence microscope. T cell receptors of helper T lymphocytes interact with an antigen-MHC-II complex in antigen-presenting cells and form immunological synapses, followed by secretory vesicle movements toward the synapse, which is visualized by fluorescent microscopy.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
