Random Priming

Random priming is a molecular biology technique that initiates nucleic acid synthesis from multiple short, randomly designed oligonucleotide primers, enabling broad representation of template sequences. In RNA workflows, these primers anneal at complementary sites across RNA molecules, and reverse transcriptase extends them to produce cDNA fragments rather than relying on a single transcript-specific primer. In cancer research, random priming supports cDNA preparation for transcriptome profiling, reverse transcription quantitative PCR, and RNA sequencing, including analyses of gene expression in tumors and model systems. Its broad sequence coverage can help detect diverse transcripts, although abundant or degraded RNA may influence representation.

SELEX protocols comprise multiple rounds of selection, each of which require regeneration of bound ligands, which in turn require fixed primer sequences flanking the random library regions. These fixed primer sequences can interfere with the selection process (false positives and negatives). Here we present a primer-free protocol.

Tractable Mammalian Cell Infections with Protozoan-primed Bacteria

This technique provides a method to harvest, normalize and quantify intracellular growth of bacterial pathogens that are pre-cultivated in natural protozoan host cells prior to infections of mammalian cells. This method can be modified to accommodate a wide variety of host cells for the priming stage as well as target cell types.
