Xpo1 Transport Regulation

XPO1 transport regulation is the control of nuclear export mediated by exportin 1, a major transport receptor that influences gene expression, cell division, and survival. In the nucleus, XPO1 binds proteins and RNA molecules carrying nuclear export signals in a Ran-GTP-dependent process, then releases its cargo in the cytoplasm after Ran-GTP hydrolysis. Cancer cells can exploit altered XPO1 activity to mislocalize tumor suppressors and other regulatory proteins, supporting proliferation and resistance to cell death. Studying this pathway has clarified cancer biology and guided development of selective XPO1 inhibitors, which aim to restore nuclear retention of growth-suppressive cargo and provide targeted therapeutic opportunities.
