Constitutional Isomers

Constitutional isomers are compounds that share the same molecular formula but differ in how their atoms are connected, making connectivity a central concept in structural chemistry. They arise when the same set of atoms can form different carbon skeletons, functional-group positions, or functional groups, producing distinct molecular structures and often different physical and chemical properties. For example, alcohols and ethers can be constitutional isomers with the same formula but different reactivity. Recognizing these relationships helps chemists interpret molecular formulas, predict reactions, analyze spectroscopic data, and design or identify compounds in organic synthesis, pharmaceuticals, and materials research.

Development of Leishmania Species Strains with Constitutive Expression of eGFP

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.

Cysteine-rich peptides fold into distinct three-dimensional structures depending on their disulfide connectivity. Targeted synthesis of individual disulfide isomers is required when buffer oxidation does not lead to the desired disulfide connectivity. The protocol deals with the selective synthesis of 3-disulfide-bonded peptides and their structural analysis using NMR and MS/MS studies.
