Tert-butyl Carbamate Protection

Tert-butyl carbamate (Boc) protection is a chemical method for temporarily masking amines during multistep synthesis, enabling other functional groups to react selectively. In this process, an amine reacts with di-tert-butyl dicarbonate to form a Boc carbamate, which withstands many basic and nucleophilic reaction conditions but can be removed under acidic conditions. Acidic deprotection restores the free amine while producing carbon dioxide and tert-butyl-derived byproducts. Boc protection is widely used in peptide synthesis, medicinal chemistry, and organic synthesis to control reaction selectivity, simplify purification, and support the sequential construction of complex molecules.
